Inflammasome sind Multiproteinkomplexe, die u. a. für die Aktivierung der proinflammatorischen Caspase-1 verantwortlich sind. Auslöser der Inflammasombildung können verschiedene danger associated molecular patterns (DAMPs) sein, die vom angeborenen Immunsystem erkannt werden und zur Aktivierung der Caspase-1 führen.
In Vorarbeiten wurden von uns enzymaktivitätsmindernde Mutationen im Pro-Caspase-1 kodierenden CASP1-Gen identifiziert, die häufiger bei Patienten mit rekurrierenden Fieberepisoden auftraten als in einer gesunden Kontrollpopulation. Die mutierten Pro-Caspase-1 Varianten induzieren über RIP2 signifikant mehr NF-kB als die wildtyp Pro-Caspase-1 und könnten so zur Inflammation beitragen. Das Ziel des Forschungsvorhabens ist daher die Aufklärung der molekularen Mechanismen, die zur Manifestation des autoinflammatorischen Phänotyps bei den Patienten führen. Insbesondere soll dabei die Rolle der mutierten Pro-caspase-1 im Rahmen des RIP2/NF-kB-Signalweges und der zytosolischen DAMP-Erkennung bei der Inflammasombildung, der nachfolgenden Transkription und Zytokinsekretion sowie der Pyroptose analysiert werden.
